1. UAB Kinome Core: (Director: Christopher D. Willey) Investigating kinase driven signal transduction cascades in a spectrum of biological systems (in vitro to in vivo), particularly related to cancer, but also other kinase driven diseases. This core utilizes a high-content peptide array platform (PamStation®12) that measures global kinase activity in cell and tissue lysates providing broad basic and translational potential. Several collaborations are ongoing across UAB campus and with other Universities and Institutes. An emphasis of the core is to develop personalized medicine approaches through evaluation of kinomic profiles in tumor specimens coupled with ex vivo profiling of small molecule inhibitors (kinase inhibitors) to guide therapy selection. Specific cancers tested include: Glioblastoma, melanoma, meningioma, lung cancer, breast cancer, head and neck cancer, leukemia, renal cell cancer, penile cancer, bladder cancer, and pancreas cancer. Several non-cancer emphases include: IgA Nephropathy, Immunology, and Schizophrenia.
2. Bioinformatics for kinomics and personalized medicine: This research collaboration involves the development of Web-based bioinformatics solutions to connect high content data, including kinomics data, with public and private “omic” databases. We are developing web-based resources (open source) for investigators to analyze kinomics data. In addition, we are developing Javascript biostatistical resources in an “app store” environment. This will allow for improved access to peer-reviewed statistical tools to work in a web-based environment. This collaboration involves Dr. Jake Chen.
3. Preclinical models and Systems Biology approaches in Glioblastoma multiforme (GBM): In collaboration with G. Yancey Gillespie, Raj Singh, and Jake Chen, we are evaluating ways to use high content data to make therapeutic decisions in GBM beginning in patient derived xenolines with the ultimate goal of doing so in patients. We have tried to miniaturize this through the use of 3D MicroTumor culture system with drug combination testing. We have created a “Proband” model of the disease and are moving towards a clinical trial paradigm.
4. MARCKS and Cancer biology as well as radiation response modulation: Investigating the protein Myristoylated Alanine Rich C-Kinase Substrate (MARCKS) in the context of tumor biology and tumor immunology. This project is focused on MARCKS impact on GBM biology. The intent is to identify the pertinent functional domain of MARCKS and to develop MARCKS targeted therapies, including small peptide therapeutics. A new direction is in the setting of tumor microenvironment and cancer stem cells.
5. Clinical/translational Research: I have particular focus in brain, lung, and head and neck cancer trials. I am a co-investigator on several clinical trials in these disease sites. In addition, I am developing collaborations with industry for translational research with patient-derived xenograft development and multi-omic testing of patient tumors. With the advent of high dose rate proton therapy, I am a member of the FLASH FORWARD research consortium evaluating the potential clinical development of FLASH therapy.
